Population-level morphological clustering and comparison based on all available specimens, together with geographic distributions, supported the recognition of at least three distinct clusters corresponding to three species in this complex: <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="franguloides">franguloides</tp:taxon-name-part></tp:taxon-name>, <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="congesta">congesta</tp:taxon-name-part></tp:taxon-name> (S.Moore) Z.Qiang Lu &amp; X.L.Zhou, and <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="caudata">caudata</tp:taxon-name-part></tp:taxon-name>. These three species and two varieties are completely or largely allopatric, and their newly defined morphological boundaries and corresponding taxonomic treatments differ substantially from those proposed previously. Sequence variation in the internal transcribed spacer (<abbrev xlink:title="internal transcribed spacer">ITS</abbrev>) region strongly supported the genetic distinction among the three species. <title>Introduction</title>
<p>Species delimitation is a fundamental task in biodiversity research (<xref ref-type="bibr" rid="B36">Wiens 2007</xref>). Over the past several decades, the proliferation of species concepts has underscored the limitations of relying on a single criterion to delimit species (<xref ref-type="bibr" rid="B11">Hey 2006</xref>; <xref ref-type="bibr" rid="B17">Liu 2016</xref>; <xref ref-type="bibr" rid="B38">Zachos 2016</xref>). A growing body of evidence indicates that single-evidence approaches can introduce systematic bias or lead to errors in species delimitation (e.g., <xref ref-type="bibr" rid="B35">Sukumaran and Knowles 2017</xref>; <xref ref-type="bibr" rid="B23">Lu et al. 2024</xref>). Different species’ concepts emphasize distinct attributes, such as morphological diagnosability, reproductive isolation, phylogenetic relationships, and ecological divergence, yet any single concept may be inadequate when applied to cryptic species, asexual lineages, recently diverged taxa, or sympatrically distributed lineages (<xref ref-type="bibr" rid="B6">de Queiroz 2007</xref>; <xref ref-type="bibr" rid="B17">Liu 2016</xref>; <xref ref-type="bibr" rid="B38">Zachos 2016</xref>). Consequently, an integrative framework that combines multiple lines of evidence is increasingly recognized as essential for robust species delimitation in taxonomic, ecological, and conservation research (<xref ref-type="bibr" rid="B6">de Queiroz 2007</xref>; <xref ref-type="bibr" rid="B36">Wiens 2007</xref>; <xref ref-type="bibr" rid="B29">Padial et al. 2010</xref>; <xref ref-type="bibr" rid="B17">Liu 2016</xref>).</p>
<p>Among the available criteria, morphological diagnosability remains one of the most practical and widely applied criteria for species delimitation (<xref ref-type="bibr" rid="B5">Dayrat 2005</xref>; <xref ref-type="bibr" rid="B35">Sukumaran and Knowles 2017</xref>; <xref ref-type="bibr" rid="B20">Lu et al. 2021</xref>). However, its reliable application requires population-level statistical comparisons, as analyses based on few specimens may confound intraspecific variation with interspecific differences (e.g., <xref ref-type="bibr" rid="B23">Lu et al. 2024</xref>). Morphological characters often vary across ontogenetic stages and during organ development (<xref ref-type="bibr" rid="B4">Crisp and Weston 1993</xref>; <xref ref-type="bibr" rid="B8">Espinosa et al. 2021</xref>); failing to account for such variation can result in substantial errors in estimates of species diversity (<xref ref-type="bibr" rid="B1">Buitrago Aristizábal et al. 2020</xref>). Moreover, hybrids frequently exhibit traits that differ from those of their parental species, and environmental heterogeneity can generate extensive phenotypic variation (<xref ref-type="bibr" rid="B19">López-Caamal et al. 2013</xref>), potentially leading to the erroneous recognition of such variants as independently evolving lineages (e.g., <xref ref-type="bibr" rid="B39">Zalewska-Gałosz et al. 2018</xref>; <xref ref-type="bibr" rid="B23">Lu et al. 2024</xref>). Therefore, species delimitation based on morphology should use specimens at comparable, morphologically stable developmental stages and be further validated using other independent lines of evidence.</p>
<p>An independent evolutionary lineage is a population or group of populations that evolves separately from other such lineages and maintains its evolutionary distinctiveness over time (<xref ref-type="bibr" rid="B6">de Queiroz 2007</xref>). Under an integrative species-delimitation framework, the recognition of a species should be supported by at least two independent lines of evidence, such as morphological differentiation, molecular divergence, reproductive isolation, or ecological differentiation (<xref ref-type="bibr" rid="B6">de Queiroz 2007</xref>; <xref ref-type="bibr" rid="B29">Padial et al. 2010</xref>; <xref ref-type="bibr" rid="B17">Liu 2016</xref>; <xref ref-type="bibr" rid="B35">Sukumaran and Knowles 2017</xref>; <xref ref-type="bibr" rid="B23">Lu et al. 2024</xref>). DNA barcoding, which uses short standardized DNA regions, provides a practical tool for identifying genetic distinction and distinguishing evolutionary lineages, particularly at the species level (<xref ref-type="bibr" rid="B10">Hebert et al. 2003</xref>). Numerous studies of plants have successfully used DNA barcodes to uncover cryptic or previously unrecognized species and to reinstate species previously placed in synonymy (<xref ref-type="bibr" rid="B12">Hu et al. 2015</xref>; <xref ref-type="bibr" rid="B34">Su et al. 2015</xref>; <xref ref-type="bibr" rid="B23">Lu et al. 2024</xref>; <xref ref-type="bibr" rid="B18">Liu et al. 2026</xref>). Thus, DNA barcodes can provide independent molecular evidence for evaluating lineages initially identified based on morphology, while geographic distribution patterns can provide additional corroborating evidence (e.g., <xref ref-type="bibr" rid="B20">Lu et al. 2021</xref>, <xref ref-type="bibr" rid="B23">2024</xref>).</p>
<p>The genus <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">Rhamnella</tp:taxon-name-part></tp:taxon-name> Miq. (<tp:taxon-name><tp:taxon-name-part taxon-name-part-type="family" reg="Rhamnaceae">Rhamnaceae</tp:taxon-name-part></tp:taxon-name>) comprises deciduous shrubs or small trees and evergreen climbing shrubs and includes 12 accepted species distributed mainly in East and Southeast Asia (<xref ref-type="bibr" rid="B31">Richardson et al. 2000</xref>; <xref ref-type="bibr" rid="B2">Chen and Schirarend 2007</xref>; <xref ref-type="bibr" rid="B9">Hauenschild et al. 2016</xref>; <xref ref-type="bibr" rid="B21">Lu and Sun 2019</xref>, <xref ref-type="bibr" rid="B22">2020</xref>; <xref ref-type="bibr" rid="B30">POWO 2026</xref>). Morphological distinctions among <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="family" reg="Rhamnaceae">Rhamnaceae</tp:taxon-name-part></tp:taxon-name> taxa are often subtle and easily overlooked (<xref ref-type="bibr" rid="B31">Richardson et al. 2000</xref>; <xref ref-type="bibr" rid="B2">Chen and Schirarend 2007</xref>; <xref ref-type="bibr" rid="B9">Hauenschild et al. 2016</xref>). Recent studies suggest that the diagnostic characters used to distinguish <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">Rhamnella</tp:taxon-name-part></tp:taxon-name> species include leaf habit (deciduous or evergreen), pubescence, petiole length, leaf length-to-width ratio, and the shape and size of dried fruits (<xref ref-type="bibr" rid="B21">Lu and Sun 2019</xref>, <xref ref-type="bibr" rid="B22">2020</xref>). Nevertheless, the morphological boundaries of several species remain unclear (<xref ref-type="bibr" rid="B2">Chen and Schirarend 2007</xref>). Morphological data were compiled from specimens accessed through <abbrev xlink:title="Global Biodiversity Information Facility">GBIF</abbrev> and <abbrev xlink:title="Chinese Virtual Herbarium">CVH</abbrev> following the methods of <xref ref-type="bibr" rid="B23">Lu et al. (2024)</xref>. All quantitative traits were measured using ImageJ v. 1.49v, whereas qualitative traits were scored based on direct observations of the specimens. Newly collected and herbarium specimens were measured and compared within and among populations. At a larger scale, the specimens were divided into four geographic regions: China, South Korea, Japan, and the Liuchiu (Ryukyu) Islands (Figs <xref ref-type="fig" rid="F1">1</xref>, <xref ref-type="fig" rid="F2">2</xref>; Table <xref ref-type="table" rid="T1">1</xref>), and morphological variation was further compared within and among regions. This approach enabled us to identify morphological variation both within and among populations or geographic regions. Morphological traits of leaves and branchlets were measured following the standardized protocols presented in Suppl. material <xref ref-type="supplementary-material" rid="S1">1</xref>: fig. The type specimens of all synonyms were also examined (Fig. <xref ref-type="fig" rid="F1">1</xref>). Specifically, we measured seven specimens that were consistent with the protologue of <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="franguloides">franguloides</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="infraspecific-rank">var.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="variety" reg="inaequilatera">inaequilatera</tp:taxon-name-part></tp:taxon-name> (<xref ref-type="bibr" rid="B28">Ohwi 1951</xref>). To ensure consistency across the morphological dataset from all examined specimens, we selected seven quantitative and two qualitative traits of leaves and branchlets that exhibited clear differentiation between populations, geographic regions or taxa for the first morphological clustering analysis (Table <xref ref-type="table" rid="T2">2</xref>). For example, lenticel density on old branchlet showed clear differentiation between populations, geographic regions or taxa, but we only obtained this dataset from 181 individuals in this complex, and all such traits were reflected by morphological comparison (Table <xref ref-type="table" rid="T4">4</xref>). Our observations indicated that petiole length was relatively stable in leaves from the middle portions of branchlets throughout development. Therefore, only leaves from these portions were measured. The second <abbrev xlink:title="Principal component analyses">PCA</abbrev> result, based on 372 fruiting specimens, recovered three major clusters (Fig. <xref ref-type="fig" rid="F4">4B</xref>). This finding differs markedly from the previous description and treatment (<xref ref-type="bibr" rid="B2">Chen and Schirarend 2007</xref>). Within Cluster C, var. inaequilatera was previously distinguished from var. franguloides by unequal-sided leaf bases, two–locular pyrenes, striate fruit surfaces, and leaf petiole length (10–15 mm) (<xref ref-type="bibr" rid="B28">Ohwi 1951</xref>). Among them, only leaf petiole length (3.8–7.6 mm vs. 7.8–15.8 mm) can distinguish the two varieties (Table <xref ref-type="table" rid="T4">4</xref>), with new variable range different from the previous description (<xref ref-type="bibr" rid="B28">Ohwi 1951</xref>).</p>
<p>Allopatric speciation is considered a common model through which reproductive isolation develops (<xref ref-type="bibr" rid="B3">Coyne and Orr 2004</xref>). Although the development of reproductive isolation and morphological discontinuities in allopatry may not be fully synchronized (<xref ref-type="bibr" rid="B37">Wu 2001</xref>; <xref ref-type="bibr" rid="B6">De Queiroz 2007</xref>; <xref ref-type="bibr" rid="B17">Liu 2016</xref>), most units exhibiting morphological discontinuities may show varying degrees of reproductive isolation (<xref ref-type="bibr" rid="B27">Mallet 2008</xref>). In addition, geographical discontinuity is itself an important isolating mechanism that maintains morphological and genetic boundaries (<xref ref-type="bibr" rid="B33">Schluter 2001</xref>). In this study, Cluster B (<tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="caudata">caudata</tp:taxon-name-part></tp:taxon-name>) is restricted to southern China (Guangdong), whereas Cluster C is distributed in South Korea, the Liuchiu (Ryukyu) Islands, and Japan (Fig. <xref ref-type="fig" rid="F5">5</xref>). Clearly, these two distinct clusters are geographically discontinuous, and no evidence suggests that the two widely recognized species should be merged into a single species. Therefore, the recognition of Cluster B (corresponding to <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="caudata">caudata</tp:taxon-name-part></tp:taxon-name>) and Cluster C (corresponding to <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="franguloides">franguloides</tp:taxon-name-part></tp:taxon-name> and its synonym from Japan) as separate species should remain unchanged (Figs <xref ref-type="fig" rid="F1">1</xref>, <xref ref-type="fig" rid="F2">2</xref>, <xref ref-type="fig" rid="F3">3</xref>, <xref ref-type="fig" rid="F4">4</xref>, <xref ref-type="fig" rid="F5">5</xref>; Table <xref ref-type="table" rid="T4">4</xref>). Furthermore, Cluster A differs considerably in morphology from Clusters B and C (Fig. <xref ref-type="fig" rid="F4">4</xref>; Table <xref ref-type="table" rid="T4">4</xref>), with B and C hypothesized to be the most closely related pair, and Cluster A is also largely or completely allopatric with respect to both (Fig. <xref ref-type="fig" rid="F5">5</xref>). Although they exhibit pronounced geographical separation (Fig. <xref ref-type="fig" rid="F5">5</xref>), there is no other new evidence to support the elevation of var. inaequilatera to species rank. Thus, the existing taxonomic treatment of the two varieties is retained.</p>
<p>Overall, our morphological delimitation of this complex differs markedly from previous taxonomic treatments in terms of taxon descriptions, circumscriptions and geographical distributions, highlighting the importance of population‐level analyses of morphological variation for delimiting species boundaries and informing taxonomic decisions.</p>

<title>Molecular evidence further supports the morphological delimitation</title>
<p>Molecular phylogenetic analysis is one of the most important approaches for inferring evolutionary relationships and delimiting species (e.g., <xref ref-type="bibr" rid="B12">Hu et al. 2015</xref>; <xref ref-type="bibr" rid="B23">Lu et al. 2024</xref>). Morphological evidence indicated that <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="caudata">caudata</tp:taxon-name-part></tp:taxon-name> and the newly-delimited <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="franguloides">franguloides</tp:taxon-name-part></tp:taxon-name> were likely the most closely related pair in this complex. However, it is also necessary to identify the key morphological characters that reflect phylogenetic relationships. Our sequence alignment revealed many variable sites that clearly distinguish the three species (Table <xref ref-type="table" rid="T5">5</xref>). Either <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="congesta">congesta</tp:taxon-name-part></tp:taxon-name> (Cluster A) or <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="caudata">caudata</tp:taxon-name-part></tp:taxon-name> (Cluster B) possessed five fixed diagnostic nucleotide sites compared to the other two species. In addition, <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="franguloides">franguloides</tp:taxon-name-part></tp:taxon-name> (Cluster C) possessed nine private sites that distinguished it from either Cluster A or Cluster B, whereas Cluster B had ten private sites when compared with either Cluster A or Cluster C (Table <xref ref-type="table" rid="T5">5</xref>). These findings suggested strong genetic divergence among these three clusters or species. Therefore, <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="caudata">caudata</tp:taxon-name-part></tp:taxon-name> (Cluster B) is one well-delimited species different from <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="franguloides">franguloides</tp:taxon-name-part></tp:taxon-name> (Cluster C), by integrating multiple lines of evidence from morphology, geographic distribution, and genetic divergence. Furthermore, the phylogenetic tree showed that <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="congesta">congesta</tp:taxon-name-part></tp:taxon-name> (Cluster A) forms an independent clade sister to the well-supported clade containing <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="caudata">caudata</tp:taxon-name-part></tp:taxon-name> (Cluster B) and <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="franguloides">franguloides</tp:taxon-name-part></tp:taxon-name> (Cluster C) (Fig. <xref ref-type="fig" rid="F6">6</xref>). This finding suggested that the absence or presence of petiole and annual branchlet pubescence, petiole length, and leaf apex length are congruent with the inferred phylogenetic relationships (Figs <xref ref-type="fig" rid="F5">5</xref>, <xref ref-type="fig" rid="F6">6</xref>; Table <xref ref-type="table" rid="T4">4</xref>). Under an integrative framework for species delimitation (<xref ref-type="bibr" rid="B12">Hu et al. 2015</xref>; <xref ref-type="bibr" rid="B17">Liu 2016</xref>; <xref ref-type="bibr" rid="B23">Lu et al. 2024</xref>), <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="congesta">congesta</tp:taxon-name-part></tp:taxon-name> (Cluster A) as a species was strongly supported by morphological and molecular evidence.</p>
<p>It should also be noted that leaf petiole length can well reflect the phylogenetic relationship (Table <xref ref-type="table" rid="T4">4</xref>; Fig. <xref ref-type="fig" rid="F6">6</xref>). The variety <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="franguloides">franguloides</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="infraspecific-rank">var.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="variety" reg="inaequilatera">inaequilatera</tp:taxon-name-part></tp:taxon-name> was originally described as a species (<xref ref-type="bibr" rid="B28">Ohwi 1951</xref>), only with the longer leaf petiole (7.8–15.8 mm) than var. franguloides (3.8–7.6 mm) (as redefined in this study, Table <xref ref-type="table" rid="T4">4</xref>). Although morphological difference among species in <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="family" reg="Rhamnaceae">Rhamnaceae</tp:taxon-name-part></tp:taxon-name> are often subtle (<xref ref-type="bibr" rid="B2">Chen and Schirarend 2007</xref>), whether this variety should be reinstated at species rank remains unclear. If both <abbrev xlink:title="internal transcribed spacer">ITS</abbrev> sequences are assigned to either variety, it is likely that var. franguloides is more closely related to <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="caudata">caudata</tp:taxon-name-part></tp:taxon-name>. However, the present study cannot determine whether the downloaded <abbrev xlink:title="internal transcribed spacer">ITS</abbrev> sequences of <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Rhamnella">R.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="franguloides">franguloides</tp:taxon-name-part></tp:taxon-name> should be assigned to both varieties or either variety (Table <xref ref-type="table" rid="T5">5</xref>). Additionally, the present study relied solely on <abbrev xlink:title="internal transcribed spacer">ITS</abbrev> marker, and sampling of Cluster C remains limited.
